Calcium Imaging
rAAV-EF1α-DIO-NES-6xHis-CaMPARI2-FLAG-HA-Myc
CaMPARI is an engineered fluorescent protein whose green-to-red photoconversion depends on simultaneous light exposure and elevated calcium, which enabled marking active neuronal populations with single-cell and subsecond resolution. CaMPARI2 is the improved sensor with brighter green and red fluorescence, faster calcium unbinding kinetics and decreased photoconversion in low calcium conditions.

Functional Gene Details
CaMPARI2.0: EricR Schreiter's team first tested the purified CaMPARI2.0 protein and found that its green and red fluorescence intensities were 60% and 130% higher than the first-generation CaMPARI protein, respectively, and the calcium dissociation rate was also increased several times, making it more sensitive. What is even more exciting is that under calcium-free conditions, the photoconversion rate (PC) of CaMPARI2.0 is significantly lower than that of its predecessor, which means that CaMPARI2.0 will show higher contrast when calcium ion concentrations are different. Using the "super chameleon" CaMPARI2.0 protein, the activities of nerves in a wide range of brain areas in complex behaviors can be studied. CaMPARI2 may have important and widespread application value in the study of brain functional atlas and connectomics. CaMPARI2 CaMPARI2-F391W CaMPARI2-L398T
