rAAV-CAG-FLEX-ArchT-EGFP-WPRE-SV40 polyA
The broad-spectrum promoter CAG regulates the expression of Cre-dependent light repressive elements. ArchT-EGFP is expressed when Cre is present, but ArchT-EGFP is not expressed when Cre is absent. The genetically modified third-generation product has unchanged excitation wavelength, improving cell membrane positioning and uniform distribution of light-sensitive tools; compared with NpHR, when the laser is turned off, Arch immediately returns from the channel open state to the closed state.

Product Details
Promoter Details
CAG, strong promoter, an artificially constructed combined promoter, composed of cytomegalovirus early enhancer and chicken β-actin promoter, 1731bp.
Functional Gene Details
ArchT: Light-driven outward hydrogen ion pump, belonging to Archaerhodopsin. Compared with Arch, although the maximum current it can form is similar to Arch, ArchT is more than three times more sensitive to light and performs well under 1–10 mW/mm2 illumination. Therefore, under the same conditions, the area that can be suppressed is about twice that of Arch. Its maximum activation wavelength is around 570nm, and it can also be activated with light around 532nm. In some neurons, concentrated firing of action potentials occurs after continuous light inhibition.
Fluorescent Tag
EGFP
Fluorescent Protein Details
EGFP: Green fluorescent protein, excitation wavelength 488nm, emission wavelength 507nm.
Virus Type
adeno-associated virus
Application
Expressing light-sensitive ion channel proteins, such as eNpHR3.0, Arch, etc., in neurons through rAAV viral vectors can specifically inhibit neurons and explore the effects of light stimulation on neurons, neural circuits and animal behavior.
Regulatory Element Details
FLEX: DIO and FLEX are very close, except for the interval 1 and interval 2 sequences used. DIO is facing inward, and Flex is facing outward.
Notes
1. Short-term storage at 4°C; long-term storage at -80°C
2. It can be stored at -80°C for more than 6 months. Each time it is frozen and thawed, the titer will be reduced by 10%, and repeated freezing and thawing should be avoided; if the virus is stored for more than 6 months, it is recommended to re-measure the virus titer before use
3. According to the experimental needs, the virus can be appropriately diluted and divided into the volume required for each experiment.
